Only you can answer that - but here's some things to consider
Setting
I'd recommend playing a game with a setting you're already fairly familiar with. For instance if you've played a lot of Warhammer try WFRP or if you've played Warmachine you might want to consider Iron Kingdoms and so on. You might be able to find an RPG based on your favourite series of books or a TV show, Comic book or Film. eg, Wheel of Time, Dr Who, Judge Dredd and Star Wars all have RPGs based on them.
Of course things are a lot more open to you if you are prepared to learn a setting from scratch, D&D has many settings to choose from including Ebberon, Krynn (DragonLance), Forgotten Realms, RaVenLoFt, Greyhawk and many others (some of which - going back to my first point - you might know from computer games like Baldur's Gate or Neverwinter Nights)
Many other RPGs also have their own settings and there are many to choose from
The final option (and the one that requires the most work) is coming up with your own setting unique to you and your group.
Experience Level
No not your character's but yours - you and your group's - have you played any RPGs before? what rule systems are you familiar with?
Are you prepared to learn a complex set of rules or do you want to start out with something a little more simple to ease yourselves into it?
if you've played D&D before and know the rules pretty well then go with a D&D or D20 System game (Pathfinder is supposed to be very good)
If your starting from scratch so far as learning RPG type rules find yourself a boxed RPG game which includes everything you need to get started - there are quite a few D&D ones out there and many other systems have starter sets too
Miniatures
Do you want to use miniatures in your RPG? - if so check the system you choose supports miniatures and also check that you have or can easily get hold of the miniatures required - you may also need to invest in Battlemats, Floor-plans and Dungeon Tiles.
D&D 3.5, D&D 4th, Iron Kingdoms, WFRP 2nd and D20 Starwars all support miniatures
WFRP 3rd, Vampire: The Masquerade and GURPS do not
